La Contamination Control Strategy (CCS) è il documento con cui l'Annex 1 tiene insieme tutti i controlli contro la contaminazione.

Il paragrafo 2.5 elenca sedici elementi da coprire: dalla progettazione dell'impianto e dal personale fino al monitoraggio, alle CAPA e al miglioramento continuo.

Tre domande che gli ispettori fanno spesso:
• è basata sul rischio o è solo un elenco di SOP?
• viene aggiornata quando qualcosa cambia?
• chi lavora in reparto sa spiegare la sua parte?

Nella Warning Letter del 22 settembre 2026 la FDA cita lo stabilimento Fresenius Kabi di Melrose Park (Illinois), che produce iniettabili sterili.

Tutto parte dalle reazioni avverse di alcuni pazienti a lotti di famotidina poi richiamati. Dietro, la FDA ha trovato:
• media fill falliti attribuiti agli operatori, senza analizzare il programma degli osservatori
• un'ispezione visiva con probabilità di rilevare particelle sotto il 70%
• indagini chiuse prima di arrivare alla causa

Quando un media fill fallisce, «errore dell'operatore» è il punto da cui partire, non quello in cui fermarsi.

A decay curve is a ventilation test.

The recovery method of ISO 14644-3 works outside cleanrooms too: raise the particle level, leave the room, log counts at short intervals and plot them on a log scale. A straight line means dilution is working and its slope is the effective air change rate, to compare with the design figure. A flattening curve means a source or supply air carrying particles. Not a compliance test. A way to turn "the air feels stale" into a number.

ISO 14644-1 defines classes 1 to 4 at 0.1 to 0.3 µm, where a 0.3 µm counter cannot classify. ISO 5 at 0.3 µm is 10,200/m³, at 0.1 µm 100,000/m³. The core of a fab needs a 0.1 µm instrument. Gowning, airlocks, subfab, incoming inspection and support-area filter scans, which are most of the floor, are ISO 6 to 8 work for a 0.3 µm handheld.
